Cytotoxic potential of biosynthesized zinc oxide nanoparticles using ceratonia siliqua l plant on mda-mb 231 cancerous cells

Studies have shown that zinc nanoparticles have potential for suppressing cancer cells, but the mechanism of action is not well defined. Zinc nanoparticles are likely to produce free radicals on theirsurface, when the nanoparticles are placed adjacent to the cell, various mechanisms in the cells are activated to minimize damage. However, if the amount of free radical production exceeds the antioxidant capacity of the cells, it can cause oxidative stress damage and ultimately death of the cell. Therefore, the present survey was carried out to examine the cytotoxic properties of nanoparticle on breast cancer cells (MDA-MB 231). The MTT examination was performed to assessment the cytotoxic capacity of the nanoparticle. MTT assay is on basis of a breakdown of salt tetrazolium by Mitochondrial enzyme of living cell and producing Violet Formazan crystals. In this method, after incubation of the cells with the various doses of nanoparticle at the desired time, the supernatant was removed and the MTT solution was added to each well and incubated for 3 hours at 37 ° C. With increasing time; incubation, the emergence of needle crystals inside the well completely was evident. Subsequently, the DMSO was added to dissolve the crystals and then the absorption rate was recorded. The result is an average of 3 times repetition presented based on ± standard deviation. Statistical analysis by ANOVA showed that nanoparticles can significantly inhibit cancer cells. As the bioavailability of cells in treatment with nanoparticles showed a significant decrease (P≤0001) compared to untreated cells. Inhibition concentrations (IC50) 24, 48 and 72 h after treatment were reported about 125, 62.5 and 31.2μg/ml. The morphological observations of cells showed significant changes in the shape and size of treated cells compared to the untreated. Thus, the healing capacity of Zn O- can open the way for further investigation of the green synthesis of nanoparticles with therapeutic properties, mainly in nanomedicine, for cancer therapy.
