Today, due to major changes in urban life and human dependency on modern conditions, physical activity has decreased, which has led to the spread of certain diseases, including diabetes, cancer,infertility. Among these, cancer is most important with high mortality rate, which has increased in last three decades. Hepatocellular carcinoma is one of the most common and deadliest types of cancers,which involved the human liver. Chemotherapy is used in order to fight against liver cancer. Because of destructive effects of chemotherapy, researchers are attracted to the nanomaterials (10-100 nm in size) which are synthesized via green methods. Among these nanomaterials, silver nanoparticles due to their anti-cancer properties have been used. In this study, we investigated the cytotoxicity effects of silver nanoparticles synthesized via green chemistry in Amaranthus cruentus seed extract on HepG2 liver cancer cells and normal HUVEC cells which determined by MTT assay. The MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) assay is cell viability assay based on effect of substances materials on different kind of cell lines including primary, normal and cancer cells.cells were treated with different concentration of nanoparticles (10, 20, 40 and 80 μg/ml) for 24 hours. After that medium has removed and MTT added to each well and incubated for 4 hours. Then, DMSO was addedin order to solve the dye. Plates were put in ELIZA reader and optical absorbance were read at 570 nm. ourresults shown that silver nanoparticles synthesized via green method in seed extract Amaranthus cruentus seed were able to inhibit HepG2 cells more potent than HUVEC cells. The silver nanoparticles inhibited liver cancer cells in the concentration of 25μg/mlwith the rate of 9.4%, while in the same concentration the rate of inhibition for normal cells was lower (11%). IC50 values for HepG2 and HUVEC cells after 24 hours treatment were 8 and 7 μg/ml, respectively. Findings of this study indicated the potential effects of silver nanoparticles synthesized via green method in Amaranthus cruentus seed extract in order to suppress liver cancer cells. however, more in vitro and also in vivo studies should perform
