Cerium oxide nanoparticles (CeO2-NPs) have shown promising approaches as therapeutic agents in biology and medical sciences. The physicochemical properties of CeO2-NPs, such as size, agglomeration status in liquid, and surface charge, play important roles in the ultimate interactions of the NPs with target cells. The purpose of this study was to examine the cytotoxic effects of CeO2-NPsbiosynthesized by Hyssopus officinalis extract (CeO2-NPs/Hf) on HepG2 liver cancer cell line. The cytotoxicity of CeO2-NPs/Hf on HepG2 cells compared to control (untreated cells) was estimated bycolorimetric method using MTT assay. This method is based on the activity of suction dehydrogenase in mitochondria living cells which turns the MTT yellow solution into Purple formazan crystals. Theamount of production of these crystals can be measured after dissolving in a DMSO with an ELISA reader. In summary 100 μl of cell suspension were grown in 96 well plates. After 24 h, the treatmentmedium containing (0, 15.6, 31.2, 62.5, 125, and 250 μg/ml) concentrations of CeO2-NPs/Hf was added on the cells and further, according to the above protocol, the bioavailability of the cells wascalculated. The data showed that nanoparticles in 250 μl concentrations had a significant inhibitory effect on liver cancer cells and the highest percentage of growth inhibition (86%) in liver cancer cellswas in this concentration and 72 h after treatment. The IC50 of 24, 48 and 72 h after exposure were 125, 60 and 15 μg/ml, respectively Biosynthesized Cerium oxide nanoparticles by Hyssopus officinalisextract have the ability to induce cytotoxicity in HepG2 liver cancer cells and they can open the door for the development of strategies in the treatment of liver cancer
