Anticancer activity of zno nanoparticle synthesized by persicaria bistorta extract against human breast cancer cells (mda)

چکیده مقاله:

Today nanotechnology is one of the innovative technologies in medical and pharmaceutical areas. Environmentally friendly metal nanoparticles, which are produced economically by green method havevarious medical applications. ZnO nanoparticle is synthesized from plants’ extracts and could exhibit high anticancer activities using its high environmental compatibility and proper nanometer dimensions.Second most frequent cause of death announced among women is breast cancer. MDA cancer cell has been found to possess various applications in nanotechnology field. In this study, the anticancer effect of ZnO nanoparticles, synthesized by plant extract (Persicaria bistorta), on the breast cancer cell-line (MDA) is analyzed.The obtained MDA was seeded in 3 plate containing 96 cells in DMEM culture with 10% serum fetal bovine and 1% penicillin /streptomycin and placed in incubator (95% humidity, 37 degree temperature, 5% co2). The cells were treated with various densities of ZnO ( 250, 125, 62.5,31.2, 15.6 μg/ml ) after 24 hours; again they placed in incubator and after (24h/48h/72h) the 3-[4,5- dimethylthiazol-2-yl]2,5-diphenyltetrazolium bromide (MTT) test was done. The result of cytotoxic assay revealed that ZnO nanoparticles has a concentration and time dependent inhibitory effect on viability of MDA cell. Inhibitory concentrations (IC50) that was obtained for nanoparticles incubation times of 24, 48 and 72 hours were about (60/60/30 μg/ml ) respectively. The perecentage viability of the treated cancer cells, after 24/48/72h, was about 56% ,55%,59% respectively. The results showed that the Zno nanoparticle had a toxic effect on the MDA cancer cell line and this finding indicate that these nanoparticles can help to develop and expand the use of nanomaterials in modern medicine for the treatment of certain diseases such as cancer
