Effect of methisoprinol on the fish rhabdoviruses (VHSV, IHNV) and herpesvirus (CyHV-3) replications and protection against viral diseases: in vitro and in vivo study in rainbow trout (Oncorhynchus mykiss) and common carp (Cyprinus carpio)

Objective: Methisoprinol (Polfa, Poland) is a synthetic compound with reported antiviral activities in human and animals. In our preliminary study, a strong inhibited effect of methisoprinol on salmonid rhabdoviruses was observed. In this investigation we examined the in vitro influence of different concentration of methisoprinol on the replication of VHSV and IHNV by measuring of viral RNA synthesis. Also we examined the influence of different doses of methisoprinol on the replication of CyHV-3 by measuring of viral DNA synthesis. Method & Materials: Monolayers of cell cultures were exposed to various doses of g/ml). The cell culture containingμmethisoprinol (0, 100, 200, 400, 600 and 800 methisoprinol were followed by infection with VHSV, IHNV or CyHV-3 suspension containing 107 TCID50 /ml. At 24 h, 48 h and 72 h after infection, cell cultures were submitted to an one-hour starvation period by replacing the medium with pre-heated PBS followed by a Ci/ml of [ 3H]-uridine or thymidine (Amersham, UK)μtwo-hour incubation with 10 and the counts per minute (cpm) evaluated in a scintillation counter (Beckman LS). Results & Conclusion: The cytopathic effects were observed in virus-infected cultures without methisoprinol (control) at 24 h after infection with CyHV-3, VHSV and IHNV. The highest percent of inhibition 72 h after g/ml (90 %), compared to theμinfection was observed at doses of 400 and 600 lowest concentration of methisoprinol where only 25 % inhibition was observed. The present study demonstrates that methisoprinol inhibits the VHSV, IHNV from rainbow trout and CyHV-3 isolated from carp. The in vivo study showed that methisoprinol significantly stimulated nonspecific cellular and humoral defence mechanisms and protection against diseases in rainbow trout and carp.
