Survey on Penaeus indicus viral diseases on hatcheries and shrimp farms in Hormozgan Province (Iran)

چکیده مقاله:

Objective: Viral diseases have become an important limiting factor for shrimp aquaculture throughout the world. Penaeus indicus is main species of shrimp farming in Hormozgan Province. P.indicus can be affected by some viral diseases as other penaeid shrimps. The important viral diseases of P. indicus include: WSSV, MBV and HPV. Recently, WSSV has emerged as one of the most serious disease problems in culture shrimp populations all over the world. MBV is a major pathogen of shrimp that harmless during the initial stages and cause mortality in PLs. Also, HPV is lethal to shrimp larvae during the first month after stocking. Method & Materials: In this study samples were collected from different hatcheries and shrimp farms that located in Hormozgan Province in 2007. A minimum of 150 PL14 and 180 shrimps were collected from each hatcheries and shrimp farms. The samples were examined by PCR and histopathological methods. From the 100 PL collected each hatcheries, 20PL were subsampled and also from 150 shrimps in each farms, 10 shrimps (gills) were subsampled for DNA extraction. For histopathological analysis for detection of MBV and HPV, 50PL and 30 shrimps preliminary were fixed in Davidson’s fixative and then sectioned and stained by hematoxyline and eosin (H&E). Results & Conclusion: The results of this survey revealed that all samples were negative for WSSV by PCR and one of hatcheries was positive to MBV and also one of shrimp farms was positive to HPV according to histopathology results. Following WSSV appearance, it spread very rapidly through most of the shrimp growing regions of Asian and the Indo-Pacific. In previous years WSSV has been detected in P. indicus in Khozestan and Busheher provinces in 2002 and 2004, but according to this study Hormozgan Province is free from this disease, now. Diagnosis of MBV is depedent upon the demonstration MBV occlusion bodies in histological preparations of the target organs or tissues (hepatopancreas and anterior midgut).In our study, prominent single and multiple, eosinophilic and spherical occlusion bodies in PL’s hepatopancreas were observed. HPV has been observed in wild and cultured penaeide shrimps in some countries. In our study, single prominent basophilic intranuclear inclusion bodies in hypertrophied nuclei of hepatopancreas was observed. Histopathological method for MBV and HPV is a standard method for diagnosis. The detection of MBV and HPV in P. indicus were for the first time in Hormozgan. There is no treatment and prevention for these diseases.
