MicroRNAs Derived of The Human Bcl2 Gene as a Novel Tool for Cancer Diagnosis and Cancer Therapy

چکیده مقاله:
Given the critical role of microRNA (miRNAs) in tumorigenesis processes, miRNAs miningand characterization could be provide promising approaches for cancer therapy, cancerdiagnosis and cancer prognostication. Bearing in mind, human B-cell lymphoma 2 (hBcl2)gene, as a member of Bcl-2 family, which act as oncogene in numerous known cancers suchas melanoma, chronic lymphocytic leukemia, breast, lung and bladder carcinoma, wasanalyzed based on in-silico survey for mining its miRNAs and their target. Detected elementswere then validated based their correlation to the putative hBcl2 mutation as well asregulatory elements. Our investigation showed that 11578 sequences with miRNAs capacityof 404 different types could be deriving from exonic (E-miRNAs) and intronic (Id-miRNAs)regions of the hBcl2 gene. However, more evaluation reveals that only 0.42% of them whichare essentially Id-miRNAs have significant validity and frequency, including hsa-mir-619-5p,has-miR-1285-5p, has-miR-5585-3p, has-miR-5096, has-miR-1273a, has-miR-1273e. On theother hand, investigation the goals of these miRNAs among 327 genes of cell cycle control,lead to disclosure ccne2, tpm3, lama4, hgf and cbl genes as the best common targets.Moreover, distribution pattern of single nucleotide polymorphism throughout the hBcl2 geneshowed significant correlation with selected miRNAs. Taken together, the results of this studywhile is confirming data about intronic regulatory function, provided a series of candidatemiRNAs, which could be used as a novel tool for cancer prognostication, diagnosis andcancer therapy.
