There are some reports on the anticancer effects of Pyrazole derivative. The goal of this studywas to examine the anti-proliferative and apoptosis-inducing characteristics of this PyrazoleTricyclic derivative. In this experimental trial, K562 cell line was cultured in the presence of100 to 0.001 mg/ml concentrations of the Pyrazole Tricyclic derivative for 24, 48 and 72 hour.Anti-proliferative and cytotoxic effects of Pyrazole Tricyclic derivative were measured bytrypan blue exclusion test and MTT assay, respectively. Apoptosis-inducing effects weremeasured by apoptotic DNA fragmentation test. Wright-Giemsa staining was used fordetermining the effects of Pyrazole Tricyclic derivative, and latex phagocytic assay was usedfor evaluating the phagocytic potential of the differentiated cells. The results showed that theK562 cells viability were sensitive to Pyrazole Tricyclic derivative. The highest apoptosis washappened in 100μg/ml at 48 hours. Encountering cells with that material caused to induceapoptosis in relation with time and concentration and our results indicated K562 cells weredifferentiated to monocyte/macrophage lineage. This finding indicated that Pyrazole Tricyclicderivative has an apoptotic-inducing effect with differentiation.
