چکیده مقاله:

Schiff base are ligands that synthesized from amino and carbonyl compounds and readilyreacts with a wide range of metal ions, and create a stable complex with the specialcharacteristics of the physical, chemical and biological. So far, significant progress was donein the use of metal complexes as a drug for the treatment of various diseases includingcancer. Previous studies have shown that the synthesized complexes of some metals havevarious effects such as anti-cancer, antibacterial and antioxidant properties. The survey wasconducted to investigate the antiproliferative and proapoptotic effects of Mn2+ complex of theN, N′-dipyridoxyl(1,2-diaminobenzene) schiff base. The MCF7 cells were exposed withvarious concentrations (0, 200, 400, 800 Âμg/ml) of desired schiff base and theirmorphological changes are examined using an inverted microscope. MTT assay wasconducted for evaluation of anti-proliferative properties of this compound and finally,occurrence apoptosis and cell cycle analysis were examined by flow cytometry. The viabilityof MCF7 cells was suppressed with dose-dependent manner (IC50 about 400 Âμg/ml).Morphological changes such as reduction in the size and volume of cells, cell deformationand shrinkage was observed in treated cells. Flow cytometry showed the percentage of cellsin the SubG1 phase significantly increased in treated cells to about 70% compared to thecontrol (%16). The Mn2+ complex of the N, N′-dipyridoxyl (1,2-diaminobenzene) schiffbase showed antiproliferative activity on MCF7 cells and induced apoptosis in these cellswhich reveals anti- cancer properties of this compound.
