Advances in recombinant DNA technology, protein and metabolic engineering, and in-silicoinvestigation lead to the use of cytotoxic products of microorganisms for anti-cancer drugsdevelopment based on targeting cell surface antigens of cancerous cells, which known asimmunotoxins. Therefore, understanding the structural and functional characteristics of thesetypes of the antigen, while lead to present a new way for cancer diagnosis, provided fittingtargets for immunotoxins designing. Considering, cancerous cell surface specific antigens ofthe breast, ovarian, uterine, colon, stomach, pancreas, thyroid, lung, liver, head and neck,testis, prostate and skin were gathered, and then corresponding nucleotide and proteinsequences as well as 3D structures of them were derived from NCBI, Uniprot and PDB,respectively. Subsequently, Motif Scan, InterProScan5 and CD Search were used forsequence characterization, and the expressions of the selected antigens were assessed viaProtegen and Proteinatlas on normal and cancerous tissues. On the other hand, thecorresponding ligands were revealed via String, and validated by Hex, Cluspro andPatchDock as docking programs. The results of this research lead to the disclosure cellsurface specific antigens including PSMA, PSA, EPCAM, GPA33, EPHA3, IGFIR, AFP, CEACAM 7, CEACAM5, CEA CAM3, CA15-3, NSE, HCG, MSLN, and MAGE4 with differentexpression on normal and cancerous tissues, and 27 key matching ligands. Meanwhile,nucleotide and protein sequence analysis of the antigens and corresponding ligands lead toprovided a series of therapeutic targets for immunotoxins designing and a profile of candidateprimers for cancer diagnosis, which ought to be more tested.
