چکیده مقاله:

Colorectal cancer (CRC) is becoming one of the most complicated challenges of humanhealthparticularly in developing countries like Iran. It is necessary to establish acomprehensive cancergenetic counseling and systematic screening program for earlydetection and to improve cancerprognosis among high risk families.Accumulating evidenceshows that enhancer of zeste homolog 2 (EZH2) is upregulatedin a broad range of cancertypes, including colon cancer Therefore, inhibiting EZH2 expressionmay be a promisingstrategy for anti cancer therapy.EZH2, as a member of the polycomb group of genes hasimportant functionsin tumor aggressiveness. Depletion of EZH2/PRC2 complex reducescolon cancercell proliferation and cell migration.the current study suggests that EZH2 has arole in the pathogenesis of CRC. Pharmacologicalinhibition of EZH2 led to significantdecrease in H3K27-3me, significant decrease in cell viability, andmigration in CRC cells. Inaddition, siRNA-mediated knockdown of EZH2 exhibited profound effectson colorectalcancer cell growth in vitro. Also,RNA interference (RNAi)-mediated intracellularEZH2depleÆŸon led to cell cycle arrest of colon carcinoma cells at the G1/S transiÆŸon. theWholegenome transcriptome analyses identified cellular genes affected by EZH2depleÆŸon in colon cancercell lines. They included several cancer-associated genes linkedto cellular proliferation or invasion,In conclusion, our results demonstrate that EZH2 depletionblocks the growth of colon cancercells[6]. The most result suggested, Influence of EZH2-siRNA transfection on the growth of SW620cells The results showed that the number ofviable cells in the EZH2-siRNA group was markedlydecreased compared tonon_siRNA.Collectively, about all results indicated , up-regulated expression of EZH2 inCRC ÆŸssues comparedwith the paired non-tumor tissues, which was related theclinicopathological significance of thepatients. upregulated EZH2 expression may contributeto the progression of the paÆŸents withCRC. A comprehensive study of epigeneÆŸcmechanisms and the relevance of EZH2 in CRC isimportant for fully understanding thisdisease and as a basis for developing new treatment optionsin paÆŸents with CRC.
