Selenoproteins have important roles in cell stress responses and redox control. GPX1 is aubiquitously expressed selenoenzyme that detoxifies hydroperoxides using reducingequivalents from glutathione. There is considerable evidence that the cytosolic form ofglutathione peroxidase 1 (GPX1) is associated with cancer risk. The aim of this study was toinvestigate the relationship between GPX1 Pro198Leu polymorphism and the risk of gastriccancer. A total of 60 patients with gastric cancer and 70 disease-free controls were recruited.Genomic DNA was extracted from blood samples. Genotyping of the GPX1 Pro198Leu wasperformed using Polymerase chain reaction-Restriction fragment length polymorphism (PCRRFLP).Among cancer cases, the distribution of Pro/Pro, Pro/Leu, and Leu/Leu genotypeswere 26.67%, 46.66%, and 26.67%, respectively. Among healthy controls, the distribution ofPro/Pro, Pro/Leu, and Leu/Leu genotypes were 41.4%, 35.7%, and 22.9%, respectively. Nosignificant difference was observed regarding the GPX1 genotype and allele frequencies atcodon 198 between cases and controls (P=0.2). The result from this study suggest that theGPX1 Pro198Leu polymorphism does not confer susceptibility to gastric cancer in the Iranianpopulation.
