Biodesulfurization of sour gas as a green and environmental-friendly procedure plays an essential role in purification of odor gas. Response surface methodology (RSM) as a quick and trustful technique was utilized to investigate the effect of independent variables on optimization of the medium composition concentration. Based on previous researches, there is a direct connection between cell growth and organism's reaction for component consumption.In order to achieve the maximum growth of isolated organism, which obtained originated from Ramsar hot spring, RSM was first employed to evaluate the best experimental conditions. To address this issue, four independent experimental parameters namely ammonium chloride, thiosulfate, yeast extract and phosphate were selected at different concentration ranges. In addition, the effect of yeast extract, sodium thiosulfate (Na2S2O3.5H2O), phosphate and ammonium chloride (NH4Cl) concentrations on cell dry weight as the dependent variable were also investigated. The maximum cell dry weight of 2.78 g/l was obtained as an optimum result, in which the composition concentration of ammonium chloride, thiosulfate, yeast extract and phosphate were 0.4, 8, 2 and 2 g/l, respectively.The effects of independent variables on interaction performance were investigated using means of the analysis of variance (ANOVA). Moreover, temperature and pH as the promising parameters on the performance of procedure was analyzed separately. The present RSM model and experimental investigation not only identified an optimum condition but also showed the interactive effects of independent variables on the bio desulfurization of sour gas.
