Rapid Detection of BRAF gene by a Silver Nanocluster DNA Probe

In this paper, a new approach for BRAF gene sequence detection was described based on thedecrease fluorescence intensity originated from oligonuclueotide-templated silver nnnanoclusters . Thespecific DNA scaffold include two fragments: one is enriched with a cytosine sequence fragment(CCTCCTTCCTCC) that can result in DNA–AgNCs formation with a high quantum yield via a chemicalreduction method, and the other is the probe fragment (5’- TATTATATTACAGAATTATAG-3’) whichcan selectively bind to its complementary single strand target . Thus, the as prepared AgNCs can exhibitdecrease in fluorescence when binding to the target DNA sequence and forming a double strand.
