چکیده مقاله:
We are moving away from trial-and-error chemistry in biomaterials. Today, the focus is on "rational design" a shift toward programming how hydrogels will behave rather than simply hoping they will perform. This review addresses recent advances in rationally engineered hydrogels that respond to single and multiple stimuli, including temperature, pH, light, enzymatic activity, glucose, and electromagnetic fields. We critically review advanced design paradigms, emphasizing the use of nanofillers such as graphene oxide, metal nanoparticles, and MXenes to enhance mechanical flexibility, electrical conductivity, and multimodal responsiveness. We also explore advanced ۴D bioprinting and machine learning technologies that will ultimately allow us to move toward patient-specific implants that evolve over time. Finally, we examine the harsh reality of translational medicine: why many promising hydrogels fail to make it from the lab to the clinic, focusing on issues such as batch-to-batch reproducibility and regulatory hurdles.
