چکیده مقاله:

In the present study, the immunogenicity of HTLV-۱ fusion epitope-loaded PLGA nanoparticles (NPs) was assessed in the presence of co-encapsulated CpG ODN adjuvant, in a mice model via subcutaneous (SC) administration. For this purpose, the multi-epitope chimera including Tax, env, and gag immunodominant HTLV-۱ epitopes was encapsulated in biodegradable PLGA NPs with CpG adjuvant. PLGA nanospheres produced by a double emulsion method had a size of <۲۰۰ nm, and encapsulation efficiency of chimera antigen was ۸۳.۹%. The release profile of radiolabeled chimera indicated that only ۲۰.۱% of chimera were released from PLGA NPs with co-encapsulated CPG ODN during one month. The nano-vaccine significantly elevated titers of IgG۱, IgG۲a, and sIgA antibodies, as well as IL-۱۰, and IFN-γ cytokines and also reduced the amount of TGF-β۱ production relative to the other formulations. The mentioned results revealed that the sustained release of chimera from PLGA as an efficient polymeric system elicited potent cell-mediated immunity without inflammatory responses against HTLV-۱.
