Rapid wound healing and functional restoration of damaged tissues remain major challenges in tissue engineering. Bioactive hydrogels based on natural polymers have emerged as promising biomaterials because of their ability to provide structural support while modulating cellular responses. In this study, the biological performance of a yeast B-glucan-based hydrogel was evaluated through in vitro biocompatibility and cell migration assays. Cytocompatibility was assessed using the MTT assay on fibroblast cells after exposure to hydrogel extracts for ۲۴ and ۴۸ h. The ability of the hydrogel to promote cell migration was investigated using a scratch wound assay, and wound closure was quantified using ImageJ software. Statistical analysis was performed using one-way analysis of variance (ANOVA). The hydrogel exhibited excellent cytocompatibility, maintaining cell viability above ۹۰% with no evidence of significant cytotoxicity. Furthermore, treatment with the hydrogel extract significantly enhanced fibroblast migration and increased the percentage of scratch closure compared with the untreated control group (p < ۰.۰۵). These findings demonstrate that the yeast ẞ-glucan-based hydrogel effectively supports fibroblast survival, proliferation, and migration, highlighting its potential as a bioactive scaffold for tissue engineering and wound healing applications.
