Polymeric nanoparticles (NPs) are versatile drug delivery platforms that offer precise control over size, morphology, and surface properties. This review examines the key polymer classes used in NP design-including poly(amino acids), polyesters, polysaccharides, poly(ethylene glycol), and vinyl polymers and evaluates conventional synthesis methods such as nanoprecipitation, self-assembly, emulsion polymerization, ionic gelation, and spray drying. While effective, these bulk methods face limitations in scalability, reproducibility, and particle uniformity. Droplet-based microfluidic technology is presented as a superior alternative, enabling the production of highly monodisperse NPs through discrete micro-reactor droplets with precise control over process parameters. Key microchannel configurations, droplet formation mechanisms, and recent applications in NPs synthesis are discussed, demonstrating that the integration of advanced polymeric NPs with microfluidic fabrication offers a promising pathway toward scalable and reproducible NPs in the drug delivery field.
