Modulatory effect of purified peptide isolated from Mesobouthus epeus Scorpion Venom on Astrocyte Cell line (۱۳۲۱N۱)

Multiple Sclerosis is the central nervous system diseases and belongs to the category of autoimmune disease. The prevalence of this disease in Iran is approaching the European level. Astrocyte cells are in nerve tissue that regulation of immune system activity by secreting various cytokines such as IL ۱۷. The aim of this study was partial purification of toxin from Mesobouthus epeus scorpion venom that has immunomodulatory effect on astrocyte cell line (۱۳۲۱N۱).In present study, purified crude venom of M.epeus scorpion. Size exclusion and reverse-phase high-performance liquid chromatography used for fractionation. Using SDS and Tricine electrophoresis, the fractional molecular weight was determined. Astrocyte cells (۱۳۲۱N۱) selected as functional cells in testing the immunomoduatory effect of venom. The viability of cells were determined by MTT and LDH assays. Astrocyte Cells activated by lipopolysaccharide, the release of interleukin-۱۷ in activated cultured astrocyte cells was estimated using ELIZA kit. Fraction ۳۳۱ (F۳۳۱) from RP-HPLC containing purified peptide with molecular weight of about ۵۵۰۰ Dalton. When activated cells were exposed to purified peptide the rate of interleukin-۱۷ release was found to be ۸۵ pg/ml which is almost similar to un-activated cells (۷۸ pg/ml). However the activated cells which were exposed to LPS but not treated with the purified peptide, IL-۱۷ increased significantly (P<۰.۰۵), to a level of ۱۴۷ pg/ml. The purified peptide (F۳۳۱) from venom of Mesobouthus epeus can inactivate the astrocyte ۱۳۲۱N۱ cells activated by LPS as indicated by decreased secretion of IL۱۷ from the cells
