چکیده مقاله:
Necroptosis is an inflammatory form of programmed cell death distinct from apoptosis and feroptosis. In contrast to apoptosis, which involves caspase activation, necroptosis is regulated by receptor-interacting protein kinase ۱ (RIPK۱) and RIPK۳, leading to the formation of the necrosome. This complex promotes cell rupture and inflammation. As a therapeutic strategy, promoting the mixed lineage kinase domain-like protein (MLKL) can increase necroptosis in cancerous tissues through phosphorylation, membrane insertion, and the formation of membrane hotspots. In cancer, necroptosis has gained significant attention as a potential therapeutic avenue, particularly as an alternative to traditional treatments similar to chemotherapy and radiotherapy. This mini-review aims to explore recent advancements in necroptosis research, focusing on novel micro- and nanoformulations that aim to regulate this pathway for cancer therapy.
