Green copper nanoparticles (g-CuNPs) were successfully synthesized using the medicinal plant Hemigraphis colorata, a tropical perennial herb belonging to the family Acanthaceae. Commonly known as ‘Murikootti’ in Kerala, this plant is renowned for its remarkable wound healing properties. It has also been reported to exhibit various pharmacological activities, including antioxidant, antimicrobial, wound-healing, and anti-anemic effects. The leaf extract of H. colorata acts as both a reducing and stabilizing agent in the green synthesis of Cu NPs. The biogenically synthesized g-CuNPs were characterized by UV-visible spectroscopy, FT-IR, XRD, SEM, TEM, and EDAX techniques. SEM and TEM micrographs confirmed the spherical morphology of the nanoparticles, with an average size of ۱۰۰ nm and a uniform distribution. The antibacterial activity of the g-CuNPs was evaluated against Staphylococcus aureus and Escherichia coli, revealing significant inhibition zones of ۱۱ and ۱۵ mm, respectively. These findings highlight their potential as antimicrobial agents against bacterial pathogens. Furthermore, the wound-healing activity of the Cu NPs was assessed in rats using an excision wound model. Wound area measurements were recorded on days ۰, ۲, ۴, ۷, ۱۴, and ۲۱ for all the groups. The progressive reduction in wound size demonstrated the ability of the g-CuNPs derived from H. colorata leaf extract to accelerate wound healing. These results suggest that green-synthesized g-CuNPs hold great promise as natural therapeutic agents for combating bacterial infections and promoting wound repair.
