Synthesis and Study of Triazoles and their Anti-Tubercular Activity against Mycobacterium tuberculosis H۳۷Rv

Tuberculosis (TB) remains a considerable global health concern, particularly due to the emergence of drug-resistant strains, necessitating the continued exploration of novel therapeutic agents. In this study, the antitubercular potential of the synthesized triazole compounds against Mycobacterium tuberculosis (MTB) H۳۷Rv was evaluated using the Microplate Alamar Blue Assay (MABA). The results demonstrated notable inhibitory activity among the tested compounds, with compound ۵B exhibiting the highest potency, with a minimum inhibitory concentration (MIC) of ۳.۱۲۵ µg/mL, compared to standard drugs such as isoniazid (MIC: ۰.۰۵ µg/mL), rifampicin (MIC: ۰.۱ µg/mL), and ethambutol (MIC: ۱.۵۶ µg/mL). Other derivatives, including ۱B (MIC: ۱۲.۵ µg/mL), ۵D (MIC: ۱۲.۵ µg/mL), and ۶C (MIC: ۱۲.۵ µg/mL), exhibited moderate inhibitory activity. These findings highlight ۵B as a promising lead candidate for further drug development. Furthermore, all the synthesized triazole derivatives were structurally characterized using Infrared Spectroscopy (IR), Nuclear Magnetic Resonance (NMR), and Mass Spectrometry (MS). Although developing effective anti-tuberculosis therapies remains challenging, the findings of this study provide valuable insights into the potential of triazole-based compounds. Overall, this study establishes a strong foundation for future investigations aimed at developing safe and effective anti-tubercular agents, ultimately contributing to the reduction of TB-related morbidity and mortality.
