Generating physiologically relevant human tissues in vitro requires precise control over the cellular microenvironment. This study presents an advanced protocol that synergies human pluripotent stem cells (hPSCs) with customized bioactive hybrid scaffolds. By leveraging the principles of directed differentiation and mechanical signaling as previously explored in foundational studies on ECM optimization we achieved enhanced structural maturity and functional consistency. Our results, validated through single-cell transcriptomics and metabolic assays, demonstrate that these engineered organoids significantly outperform standard ۳D models. This work provides a scalable, high-fidelity platform for drug discovery and personalized regenerative medicine.
