Background and Aim: Spexin is a novel adipokine linked to various metabolic effects, including insulin and glucose homeostasis. This study aimed to investigate the impact of ۸ weeks of high-intensity interval training (HIIT) on spexin gene expression levels in adipose tissue and insulin resistance in the skeletal muscle tissue of obese female rats with type ۲ diabetes (T۲DM).
Methodology: For this research, ۴۰ eight-week-old female Wistar rats, with an average weight of ۲۰۰g, were acquired from the animal farm of Kerman University of Medical Sciences for laboratory use. After acclimatization to the new environment, the ۴۰ rats were divided into four groups of ۱۰: (۱) healthy control (non-diabetic, no training), (۲) diabetic control (diabetic, no training), (۳) healthy trained (non-diabetic, trained), and (۴) diabetic trained (diabetic, trained). The diabetic control and diabetic trained groups were induced with T۲DM and then subjected to an eight-week training intervention. Diabetes was induced by a single intraperitoneal injection of ۳۵ mg/kg streptozotocin (STZ). HIIT was performed three sessions per week for eight weeks. Data analysis involved a two-way ANOVA, and Tukey's post-hoc test was used for inter-group comparisons.
Findings: The results showed that eight weeks of high-intensity interval training led to a significant increase in insulin levels, a decrease in insulin resistance, and an increase in spexin expression in adipose tissue and spexin levels in serum.
Conclusion: Overall, it can be concluded that eight weeks of high-intensity interval training increased the expression of genes related to fat and glucose metabolism, such as spexin in adipose tissue and serum. This indicates increased energy expenditure and improved metabolic function in type ۲ diabetic conditions. These findings suggest that high-intensity interval training can be an effective strategy for improving the metabolic and hormonal status in individuals with type ۲ diabetes and obesity.
