Background: Diarrhea in calves play a major role in economic losses of farms. The present study was performed to analyze the presence of BVDV in calves in autumn and winter in three provinces of Iran. Methods: A total of ۵۰ diarrheic fecal samples were collected from calves under one-month age of Holstein dairy cattle herds of some provinces of Iran. The samples were directed to antigen detection by RNA extraction, and cDNA synthesis for PCR amplification. Two sets of primers framed two different regions within the gene coding for protein p۸۰, the third set of primers was selected to amplify cDNA within the envelope glycoprotein (gp۵۳) region. Results: Out of ۵۰ calves’ diarrheic fecal samples, ۷ (۱۴%) were positive for BVDV RT-PCR. The distribution of the BVDV-positive samples among bovine diarrheic calves was ۶, ۲, and ۶% in Tehran, Alborz and Qazvin, respectively. Conclusion: RT-PCR indicated the prevalence rate of BVDV in autumn and winter in Iran. The study showed that positive cases detected by RT-PCR were in line with the previous studies regarding to the result of RT-PCR and the prevalence of BVDV. RT-PCR is a rapid, sensitive and adaptable method for detecting pestivirus nucleic acids. Further molecular and serological studies are needed to address the role of BVDV as a cause of diarrhea in calves.
