Background: Mycoplasmosis is one of the important diseases of poultry that cause great economic losses in the poultry industry. Mycoplasma gallisepticum is the most important poultry mycoplasma pathogen, which is commonly causes chronic respiratory disease in chickens. Achieving rapid, accurate, and definitive diagnosis of mycoplasma is great importance to health officials and veterinarians of broiler breeder farms, given its economic importance, because early and sensitive diagnosis can help farm health management maintain the general health of the herd, prevent disease progression, and select appropriate treatment policies. Currently, two serological methods, rapid serum agglutination (RSA) and Enzyme-Linked ImmunoSorbent Assay (ELISA) are used to screening Mycoplasma gallisepticum in industrial poultry. The aim of this study was serological screening of Mycoplasma gallisepticum in broiler breeder farms in different provinces of Iran. Methods: For this purpose, ۳۴۴۱ blood samples were randomly collected from a total of ۱۰۸ broiler breeder flocks from different provinces of Iran. The samples were immediately sent to the laboratory and after separating the serum from the samples, the rapid serum agglutination test (RSA) in accordance with existing standards was used to monitor the level of infection. Results: From total ۳۴۴۱ samples from ۱۰۸ different flocks, ۱۹ samples (۰.۵۵%) from seven flocks (۶.۴۸%) were Mycoplasma gallisepticum positive in the RSA test. Conclusion: Given the report of infection of breeder broiler farms with Mycoplasma gallisepticum, the need to implement biosecurity guidelines is felt. Also, due to the high sensitivity of the RSA test and the possibility of false positive results, it is necessary to perform complementary tests such as ELISA.
