Background: The treatment of infected open wounds poses a formidable challenge, leading to impaired wound healing. In this study, the effects of two nanocomposites, gold/perlite (GPNCT) and gold/bohemite (GBNCT), which possess antibacterial and antioxidant properties and have been widely used for various therapeutic purposes, were compared in terms of their ability to enhance wound healing. This study aimed to evaluate the effects of topical application of ointments prepared from GPNCT and GBNCT on excisional infected wound healing in a mouse model. Methods: ۳۶ healthy male mice were used in this experiment. A single circular full-thickness wound was created on the dorsal surface of each mouse. An aliquot containing ۲.۵ × ۱۰⁶ CFU of Staphylococcus aureus (StAu) and Pseudomonas aeruginosa (PsAe) inoculated into the wound. The animals were then divided into four groups: control, mupirocin®, and GPNCT and GBNCT ۲% (w/w) ointments, respectively. For this purpose, in vitro antibacterial activity and in vivo wound size were measured. Results: MIC and MBC values for GBNCT were ۶.۳۳ ± ۰.۵۱ μg/mL and ۵.۳۰ ± ۰.۳۳ μg/mL for StAu, and ۶.۴۵ ± ۰.۷۳ μg/mL and ۵.۸۵ ± ۰.۷۹ μg/mL for PsAe. For GPNCT, these values were ۶.۹۰ ± ۰.۵۰ μg/mL and ۶.۰۰ ± ۰.۹۲ μg/mL for StAu, and ۷.۲۰ ± ۰.۵۱ μg/mL and ۶.۱۰ ± ۰.۹۹ μg/mL for PsAe. Topical application of GPNCT and GBNCT significantly (p < ۰.۰۵) decreased wound area and total tissue bacterial count. Among the two nanocomposites, GBNCT demonstrated superior effects on the rate of infected wound healing. Conclusion: In summary, it can be concluded that GBNCT nanocomposite can accelerate the healing of infected wounds through its antibacterial properties. Future studies should investigate long-term degradation, immune responses, and the scalability of this treatment for clinical applications.
