This research is dedicated to investigating the bioactive compounds of the Paraboea leuserensis B.L. Burtt ethyl acetate fraction as potential beta-secretase ۱ (BACE۱) inhibitors, a central enzyme in amyloid beta (Aβ) peptide production that oligomerizes to create neurotoxic Alzheimer's disease plaques. Since BACE۱ is a central enzyme in the amyloidogenic pathway, enzyme inhibition presents a promising therapeutic approach to delaying or even preventing the development Alzheimer's disease. Phytochemical analysis of the extract provides evidence of high phenolic acid and flavonoid content, two classes of secondary metabolites well known for their neuroprotective potential. In vitro assays illustrate that the extract significantly suppresses the activity of BACE۱, a marker that renders the natural product pharmacologically active as a preventive agent for neurodegenerative disorders. To further improve the bioavailability and effectiveness of such bioactive molecules, an alternative nanophytotherapy strategy based on nanoparticle formulation has also been explored. The work here opens up new avenues to explore the use of Paraboea leuserensis as a source of a therapeutic candidate and forms the basis for future studies on identifying active constituents and determining molecular mechanisms of action.
