In this investigation, the primary objective was to synthesize and design a drug delivery nanostructure utilizing a multilayer hydrogel framework composed of natural polymers, specifically ethyl cellulose, carboxymethyl chitosan, and hyaluronic acid. The polymeric composition was structured such that the inner layer consisted of ethyl cellulose, the middle layer incorporated hyaluronic acid, and the outer layer was formed from carboxymethyl chitosan, which included the drug and gold nanoparticles. Initially, individual solutions of each polymer were prepared under specific thermal conditions and magnetic stirring speeds. Subsequently, these polymer solutions were sequentially deposited in Petri dishes to create successive layers. In the final phase, a solution of carboxymethyl chitosan containing the drug paclitaxel and gold nanoparticles at four concentrations (۰, ۲.۵, ۵, and ۷.۵ wt%) was applied as the outer layer. Following the placement of samples in a freezer, a freeze-drying process was conducted at -۴۲°C and +۰.۱ bar pressure for ۴۸ hours. To assess the characteristics of the synthesized nanostructure, a variety of tests, including X-Ray Diffraction (XRD), Scanning Electron Microscopy (SEM), as well as mechanical, porosity, and drug release evaluations, will be performed. Furthermore, to evaluate the anticancer efficacy of the scaffold, cancer cell culture experiments will be conducted in the presence of the synthesized nanofibers. The results of these investigations are anticipated to demonstrate the potential application of these nanostructures in targeted drug delivery systems for cancer therapy. This study aims to establish a novel drug delivery system based on multilayer hydrogels composed of natural polymers combined with gold nanoparticles and anticancer agents to enhance the efficacy of cancer treatment. The results from  this research may contribute to the development of more targeted and effective DDS for the treatment of various malignancies.
