In this study, the phenomenon of fluorescence resonance energy transfer (FRET) between nicotinamide (NAM) and bovine serum albumin (BSA) protein was investigated. To this end, the interaction of nicotinamide (the acceptor) and bovine serum albumin (the donor) was investigated under pseudophysiological conditions using ultraviolet-visible (UV-Vis) and fluorescence spectroscopy. Fluorescence resonance energy transfer can be an accurate measure of molecular proximity at angstrom distances (۱۰–۱۰۰ angstroms) and is most efficient when the donor and acceptor are within the Förster radius (R₀), the distance at which ۵۰% of the excitation energy is transferred to the acceptor, which typically ranges from ۳ to ۶ nm. In this system, the calculated distance between the donor and acceptor (r) was ۳.۳۵ nm, which is less than the critical limit of ۸ nm and indicates a high probability of energy transfer. The energy transfer efficiency was calculated to be ۲۰.۲%. Also, the fact that r > R₀ (۲.۶۷ nm) confirms the existence of a static quenching mechanism in the NAM-BSA interaction.
