Wound Healing Enhancement Using Fermented Nano Coffee Extract (Coffea canephora)

Burns arise from tissue damage caused by various agents, including chemicals, electricity, heat, sunlight, and even nuclear radiation. This study investigates the potential of Robusta coffee, a natural ingredient readily available in Indonesia, to facilitate burn wound healing. Robusta coffee (Coffea canephora) possesses flavonoid compounds with potential wound-healing properties. Fermentation of C. canephora using Saccharomyces cerevisiae can enhance the concentration of bioactive compounds. This study examines the efficacy of second-degree burn wound healing in male Mus musculus Balb/c mice using topically applied nano-formulated preparations derived from fermented C. canephora. Nano fermentation of C. canephora is an extract of C. canephora prepared with carbomer, polyethylene glycol, and triethanolamine matrix, using the ultrasonic method. The fermented C. canephora nano was characterized using UV-Vis spectroscopy, FTIR, DLS, and AFM. Nano fermented C. canephora (Cof-NPs) showed a high loading efficiency of ۹۸.۴۷%, loading amount of ۴۹.۲۳%, and contained ۹.۱۵ mg/g caffeine. The test used ۲۴ M. musculus, which were divided into six groups; five groups were burned and treated twice a day. To evaluate its potential in burn wound healing, histological examination of skin tissue was conducted. Specifically, a ۲۰% dose of Cof-NPs caused complete re-epithelialization of M. musculus wounds, accompanied by a significant increase in collagen content and a decrease in the number of inflammatory cells by ۴۵.۶۸% and ۱۱۵.۷۵ cells/mm۲. This suggests that Cof-NPs can promote wound healing, especially in burn wounds. These findings highlight the potential of nano-fermented C. canephora as an effective therapeutic agent for natural-based burns.
