Characterization of Serum Concentration of CD۸, CD۴, Interfron-γ, and Interleukin-۴ in ۴T۱ Tumor Inoculated Mouse Model

Background: Malignancies in domesticated animals, including cats and dogs, present significant health challenges. The ۴T۱ tumor-inoculated mouse model is pivotal for studying tumor biology and therapeutic strategies.Objectives: This study aimed to characterize immune system alterations in the ۴T۱ tumor-inoculated mouse model by assessing serum concentrations of CD۴ and CD۸ antigens, along with cytokines interferon (IFN)-γ and interleukin (IL)-۴.Methods: BALB/c female mice were subcutaneously inoculated with ۴T۱ cells. Serum levels of CD۴, CD۸, IFN-γ, and IL-۴ were quantified using ELISA, and histopathological analysis was performed to confirm tumorgenesis.Results: Tumor-bearing mice exhibited significantly elevated CD۸ levels (mean: ۷.۲۶۱۳ ng/mL) compared to healthy controls (mean: ۱.۹۷۴۹ ng/mL), whereas CD۴ levels were lower in tumor-bearing mice (mean: ۲.۷۶۳۲ ng/mL) compared to controls (mean: ۴.۸۶۷۷ ng/mL). IFN-γ levels were reduced (mean: ۱۰.۹۵۲۳۸ vs ۲۲.۸۵۷۱۴ pg/mL), while IL-۴ levels were increased (mean: ۲۷.۷۶۹۴۲ vs ۱۰.۸۵۴۵۲ pg/mL).Conclusion: The ۴T۱ tumor model induces significant immune alterations, highlighting its utility for studying tumor immune evasion mechanisms and advancing immunotherapy strategies.
