Bioactivity of Nano 𝑪𝒖𝒓𝒄𝒖𝒎𝒂 𝒍𝒐𝒏𝒈𝒂 Encapsulated in Carboxymethyl Chitosan

Nanoencapsulation of C. longa extract with carboxymethyl chitosan (CMCs) was formulated to prevent its bioactive deterioration in anti-dengue and anti-cancer activities. The particle size of C۱-NPs was measured at ۴۶۲.۵ ± ۱۰.۸۷ nm with a PDI value of ۰.۲۸ ± ۰.۰۵ which indicates homogeneous particle size distribution. C۱-NPs were observed to be stable at ۳۰-۱۰۰ °C but less stable at high pH and salt concentrations. C۱-NPs had a %LE of ۱۹.۷۷ ± ۷.۴۶% and %LC of ۳۹.۵۴ ± ۱۴.۹۱%. The release of C۱-NPs at pH ۲, was observed at ۲.۸۹ ± ۰.۳۳%; at pH ۷, it was ۸.۵۰ ± ۰.۹۸%; and at pH ۸.۵, it was ۵.۸۹ ± ۰.۴۰%. The best release was observed at pH ۷. Anticancer activity against the MCF-۷ cell line showed that C۱-NPs had better inhibitory activity compared to C. longa. C۱-NPs had an EC۵۰ value of ۴.۷۵ ± ۰.۴۲ µg/mL, while C. longa had ۳۳.۰۳ ± ۷.۲۰ µg/mL. C. longa extract and C۱-NPs exhibited anti-dengue activity with EC۵۰ values of ۵۴.۰۹ ± ۱۱.۸۴ and ۹.۰۲ ± ۰.۰۰ µg/mL, respectively. Based on the selectivity index (SI), C۱-NPs are more selective than the C. longa extract. Proven to improve bioavailability and controlled release, C. longa extract nanoencapsulation enables precise targeting of bioactive compounds to enhance bioactivity.
