Background: Sperm cryopreservation is associated with a decline in quality, primarily due to the excessive production of reactive oxygen species (ROS) and physical-chemical changes during the freezing and thawing process. Numerous studies have demonstrated that the use of antioxidants in the cryopreservation medium can mitigate the damage caused by this process. The aim of this research was to investigate the effects of pre-treatment with thymoquinone at concentrations of ۰, ۲۰, ۵۰, and ۱۰۰ micromolar on human sperm parameters after freezing and thawing process. Methods: Semen samples were collected from twenty-five normozoospermic men. Each sample was divided into five equal aliquots: one fresh control group and four frozen-thawed groups supplemented with Thymoquinone at concentrations of ۲۰, ۲۰, ۵۰, and ۱۰۰uM. Pre-cryopreservation and post-thaw assessments were conducted to evaluate sperm quality parameters, including sperm motility, vitality, DNA fragmentation and fertility potential, marked by levels of ADD۱. Result: Cryopreservation significantly reduced sperm quality. Thymoquinone significantly increased total and progressive motility, as well as sperm vitality at all three doses, with the highest effects observed in the ۱۰۰ uM group (p<۰.۰۱). the application of ۱۰۰ uM TQ resulted
