This study explores how blood clotting factors VII, VIII, and IX bind to adenovirus type ۵, which is known to interact with receptors such as CAR, HSPG, and coagulation factor X. While Factor X binds to Ad۵ through electrostatic interactions to the hexon protein, the potential for other clotting factors to bind and impact viral targeting remains uncertain. Using molecular modeling and bioinformatics techniques, we examined these interactions to better understand their role in directing Ad۵ to the liver. Zeta potential measurements revealed that binding of factors VII and VIII reduced the negative charge of the virus, whereas factor IX caused only a slight change. Zeta sizer analyses showed larger increases in intensity and particle number for complexes with factors VII and VIII, with volume-based measurements indicating higher levels for factors VIII and IX. Protein docking simulations suggested that factors VII and VIII are positioned near the adenovirus fiber, indicating possible interactions, while factor IX appears unlikely to bind. In summary, adenovirus ۵ may associate with factors VII and VIII, but its interaction with factor IX remains inconclusive.
