Cancer poses a significant global health burden and requires innovative therapeutic strategies. Janus nanoparticles, distinguished by their unique anisotropic architecture and dual or multiphase characteristics, have gained recognition as potential candidates in the field of cancer therapy. This review presents an extensive overview of Janus nanoparticles, emphasizing their distinct benefits and varied applications in the treatment of cancer. It primarily focuses on their application in targeted delivery systems through which they can use their segregated compartments to simultaneously deliver multiple therapeutic agents to a tumor site to reduce off-target effects in cancer therapy. Furthermore, it addresses the role of Janus nanoparticles in cancer imaging, theranostics, and emerging therapeutic strategies. The idea is to design multifunctional nanoparticles that combine imaging and therapy on a single platform, thereby combining different therapeutic modalities. Ultimately, this article discusses the challenges involved and the future prospects with respect to Janus nanoparticles for cancer therapy and highlights the possibilities they hold to change the landscape of cancer therapeutics.
