Introduction: Wound healing is a physiological and complex process that can be stopped for any reason, leading to chronic infections and prolonged healing. Human attention has been drawn to the use of natural extracts, especially sea creatures such as echinoderms. This attention is due to their important therapeutic properties. This study was conducted with the aim of investigating the effect of methanol extract of sea cucumber on wound healing in rats. In previous research, many bioactive compounds have shown promising potential in the repair and regeneration of various body tissues, including the skin, making the extract of the sea cucumber Holothuria leucospilota promising options to become a tool in wound management and treatment. Methods: In this experimental study, methanolic extract of Holothuria leucospilota was prepared from individuals collected at a depth of ۱۰-۱۵ meters of the Suza coast of the Persian Gulf in Qeshm Island. Two concentrations of extract (۰.۵% and ۱%) were formulated into an ointment base cream. Forty male Wistar rats averaging ۱۸۵ ± ۵ grams in weight were divided into four groups; a control group, a sham group treated with base cream, and two treatment groups with remedial ointments containing ۰.۵% or ۱% extract. Wounds of ۶±۱ mm were created on the dorsal skin of the Wistar rats using punch biopsies. For ۱۳ consecutive days, rats received ۱ ml of treatment either of the base cream, or each extract based ointment. Wound healing was assessed and measured on day ۰, before treatment, and on days ۴, ۷, ۱۰, and ۱۳ with regard to size of the diameter of the wound Results: The results indicate that both treatment groups showed significant improvement in wound healing over the control and sham groups. Also, the ۱% treatment group greatly reduced the diameter of wounds at all measurement intervals, from the beginning to days ۷, ۱۰, and ۱۳ (P
