چکیده مقاله:

A۵۴۹ is a human lung adenocarcinoma cell line known for its KRAS mutation and is frequently used as a model for type II alveolar cells and non-small cell lung adenocarcinoma. The INK۴-ARF locus, located in the ۹p۲۱.۳ chromosomal region, contains a gene cluster that includes P۱۴ARF, P۱۶INK۴A, P۱۵INK۴B, and the long non-coding RNA ANRIL/CDKN۲B-AS۱, which is transcribed in the antisense direction of CDKN۲B. Structurally, a shared bidirectional promoter regulates the transcription of ANRIL and P۱۴ARF. P۱۶INK۴A is positioned near the first exon of ANRIL, while P۱۵INK۴B is located within the first intron of ANRIL, also in the antisense direction. P۱۶INK۴A, P۱۴ARF, and P۱۵INK۴B act as tumor suppressors and negative regulators of the cell cycle; their deletion has been reported in the A۵۴۹ cell line. ANRIL functions as a negative regulator of these genes and has an oncogenic role in various cancers. To date, ANRIL deletion in the A۵۴۹ cell line has not been reported.
