Synthesis And Characterization of Insulin-MOF-۵ Loaded into Cellulose Nanocrystalline-Based Dissolving Microneedle as Anti-Diabetic Delivery System

Dysregulation of insulin metabolism in pancreatic β-cells necessitates using exogenous insulin injections for individuals with diabetes mellitus (DM). However, such injections are frequently associated with some challenges, such as hypoglycaemic episodes and physical discomfort. The aim of this study was to design a novel insulin delivery platform via the smart material Metal-Organic Framework-۵ (MOF-۵), incorporated into Dissolving Microneedles (DMN), as a more effective and less invasive alternative. In this regard, DMN fabrication employs cellulose nanocrystals (CNC) derived from modified cellulose from bagasse biomass. The findings of the present study revealed that X-ray diffraction (XRD) analysis confirms the successful synthesis of CNC, with a crystallinity index of ۵۷%. The incorporation of MOF-۵, characterized as a porous and responsive material, significantly enhances insulin delivery efficiency. Scanning electron microscopy-energy dispersive X-ray spectroscopy (SEM-EDX) confirms the development of pore structures of MOF-۵, with morphology optimized for microneedle applications. Furthermore, XRD analysis of MOF-۵ indicates a crystallinity index of ۶۴%, reflecting its structural integrity. MOF-۵ serves as a release-modulating agent, ensuring sustained insulin administration and mitigating the risk of excessive release. Integrating DMN with MOF-۵ provides a highly efficient and minimally invasive insulin delivery method for diabetes management. In vitro experiments demonstrate a controlled insulin release of ۷۸% over ۸ hours, while in vivo studies suggest gradual and controlled blood glucose regulation in animal models using the MOF-Ins formulation.
