Effects of Persian Shallot Extract on Inhibition of Adipogenesis in ۳T۳-L۱ Cells

Today, the industrial world has encountered the problem of obesity which can be a risk factor for diseases such as cancer, cardiovascular diseases, and type ۲ diabetes. Various strategies have been proposed for the management of obesity, one of which includes the utilization of medicinal plants. This study examines the impact of hydroalcoholic extract from Persian shallots on the adipogenesis of the ۳T۳-L۱ cell line. The cell viability of Persian shallot extract was evaluated using the MTT assay following treatment with various concentrations of the extract, with assessments conducted at ۲۴-hour and ۴۸-hour intervals. The lipid content of the cells was evaluated through Oil-Red O staining, in conjunction with quantitative real-time PCR analysis to assess the expression of the PPARγ gene. The results from the MTT test indicated that concentrations below ۲۵۰ μg/ml maintain cell viability levels exceeding ۷۰%. However, the concentration of ۱۲۵ μg/ml was also employed in this context. Upon reaching ۸۰% confluency, the cells are treated with a differentiation medium for two days. The subsequent step involved transitioning to an expansion medium for six days. The expansion medium was refreshed every ۴۸ hours during this period. During the differentiation process on Day ۸, we observed the presence of lipid droplets that had accumulated. In the Oil-Red O staining analysis, our observations indicated a significant reduction in the lipid content of mature adipocytes treated with ۱۲۵ μg/ml compared to the control group of mature adipocytes. The gene expression analysis revealed a significant decrease in PPARγ expression on the eighth day of differentiation, with a reduction of ۷۷% compared to the control group (P value = ۰.۰۲۸). These findings indicate that Persian shallot may represent a viable treatment option for managing obesity.
