Dalbergia stipulacea is a medicinal plant with a long history of traditional use. Recent scientific investigations have focused on its potential as a source of novel analgesic and hypoglycemic agents.To evaluate the plant’s therapeutic potential, in vivo and in silico studies were conducted. Methanolic extracts of D. stipulacea were administered to mice to assess their analgesic and hypoglycemic effects. A study utilizing in-silico molecular docking and ADME was carried out to evaluate the oral bioavailability and binding affinity of the compounds from Dalbergia stipulacea. Methanolic extracts (ME) were used in the study at doses of ۲۰۰ mg/kg and ۴۰۰ mg/kg to evaluate the analgesic and hypoglycemic effects. In the acetic acid-induced writhing test, ME (۲۰۰ mg/kg) and ME (۴۰۰ mg/kg) demonstrated significant results, scoring ۴۶.۴% and ۵۶.۸% of inhibition respectively. In the tail immersion test, ME at a ۴۰۰ mg/kg dosage resulted in the largest delay time increase of ۱۴.۷۶ ± ۰.۶۸ at ۱۲۰ minutes. A substantial reduction in plasma glucose levels was noted in the streptozotocin-induced hypoglycemia test for ۲۰۰ mg/kg and ۴۰۰ mg/kg of methanolic extract.In-silico studies utilizing molecular docking have further elucidated the mechanisms underlying these effects. The in-silico investigation revealed that Dalpulapan D & R, R-velucarpin A had the best docking score of around -۱۰.۸ kcal/mol against Cox۲, as compared with diclofenac's -۷.۲ kcal/mol. However, as compared to Glibenclamide (-۹.۲ kcal/mol), R-velucarpin C demonstrated the highest docking score (-۱۰.۹ kcal/mol) against the sulfonylurea receptor.
