Elevated Serum C-Reactive Protein in Female Periodontitis Model Induced by Porphyromonas gingivalis: In Vivo and Molecular Docking

Periodontitis presented a close relationship with various systemic conditions. The key to the development of periodontitis is P. gingivalis. C-reactive protein (CRP) is used as one of the main indicators of systemic inflammation that caused by bacteria infection. This study focused on a female rat model induced by P. gingivalis to evaluate the increase in serum CRP levels through in vivo and molecular docking approaches. This study used female animal models that induced by P. gingivalis suspension in the mesial gingival sulcus of the first right and left molars of the lower jaw with a dose of ۰.۰۵ ml, every three days for ۱۹ days. The euthanasia procedure was carried out chemically using ketamine at a lethal dose, based on the period of observasion and blood was taken intracardially. CRP measurement used an ELISA kit. Molecular docking analysis used protein Data Bank website, Pymol software, and ClusPro ۲.CRP level was statistically analysed by analysis of variance (ANOVA) (p<۰.۰۵). The result indicated that serum CRP levels in the female periodontitis rat model group were significantly higher than the control group (p=۰.۰۰۰). There was no significant difference in the groups within the observation period range (p>۰.۰۵). However, there was a difference in serum CRP levels between the control group and the female periodontitis model in the same observation period (p<۰.۰۵). The CRP- Lysine-specific gingipain (KGP) bond energy was the lowest and most stable among the CRP bonds with other P. gingivalis virulence factors (-۱۰۳۸.۶). Briefly, P. gingivalis induction increases CRP levels in a female periodontitis model.
