Chitin-glucan complex (CGC) is a major structural component of yeast and fungi cell wall. In this research, three types of pretreatment methods namely, low power ultrasonic bath, high power ultrasonic and autoclave, were used for CGC extraction from the yeast Saccharomyces cerevisiae. The yield of each extraction condition was measured and the effect of pretreatment conditions on the structural properties of the extracted CGC was studied by moisture content, FTIR and CHNOS. An inverse relation between the yield and moisture content of the samples was found. FTIR spectra showed slight changes in the bands compared to the initial yeast, however, the wavenumbers of the bands among the samples with different pretreatment methods was not changed significantly. FTIR spectra confirmed the presence of chitin and beta glucan in the extracted CGC samples. Besides, FTIR spectra confirm that the chemical structure of CGC was not affected by the pretreatment. Based on CHNOS results, autoclaving for ۴۰ min resulted in the highest C/N ratio among the pretreatment methods, and high power ultrasonication reached the lowest C/N ratio which is an indication of higher chitin content. The antioxidant activity of the obtained CGC samples was assessed by DPPH radical scavenging method. All of the extracted CGC samples showed free radical scavenging activity in the range ۵۳-۵۸%. Free radicals are one of the main causes of skin aging. Accordingly, it can be concluded that CGC samples obtained from Saccharomyces cerevisiae can be considered as a promising anti-aging ingredient for skin health applications.
