Temporal lobe epilepsy was considered as an incurable disease with not clearly defined mechanisms. Ferulic acid (FA) have shown anti-inflammatory, antioxidant and neuroprotective potentials. The aim of this study is evaluating the effect of FA on kainic acid (KA)-induced seizure, and a well known neuroinflammatory and oxidative stress mediator in hippocampus of mice. Materials & Methods: Sixty male mice (۲۵ to ۳۰ g) were divided into ۶ groups. Control: received normal saline (NS:۱ ml/kg body weight, intraperitoneally (i.p)), KA (۱۰ mg/kg, i.p), FA (۲۰ and ۸۰ mg/kg, i.p) daily for ۷ days, FA (۲۰ and ۸۰ mg/kg, i.p) + KA (۱۰ mg/kg, i.p) daily for ۷ days. Animal Convulsive behaviors were recorded up to ۲ hours after KA injection. Further, interleukin ۱beta (IL-۱β) and carbonyl protein (CP) levels were determined in homogenate of mice hippocampus immediately after behavioral tests. Results: The injection of KA led to seizure and increased IL-۱ β and CP content in mice hippocampus. Pretreatment with FA at both doses suppressed the seizure and associated increase in hippocampal IL-۱β and CP in mice. Conclusion: Our results suggest that pretreatment with FA not only inhibited KA-induced seizures, but also suppressed the seizure targeted effects on IL-۱β and CP production in the hippocampus. Therefore, FA can protect temporal lobe epilepsy in mice by modulating inflammatory factors and oxidative stress in the hippocampus.
