Expression level of long non-coding RNAs MALAT, PVT۱, and HOTAIR and their targets in cervical cancer cells treated with Coumarin plus Oxaliplatin

Cervical cancer is a grave and challenging cancer to overcome. A possibly valuable factor in predicting the outcome of this type of cancer is the existence of long non-coding RNAs (lncRNAs), as they have been shown to influence several mechanisms involved in the advancement and spread of cancer. Therefore, lncRNAs may have value in predicting the outcome of cervical cancer. In this way, a natural substance with some beneficial anti-cancer qualities is Coumarin. Method and Materials: Throughout our investigation, we provided administration of either Coumarin or Oxaliplatin separately or in tandem to Hela cells. Hela cells were subjected to either Coumarin, Oxaliplatin, or a combination of both in this inquiry. Our analysis focused on examining the expression of distinct long non-coding RNAs (lncRNAs), such as MALAT, PVT۱, and HOTAIR, and their corresponding targets, including miR-۱۹۵, miR-۲۰۰b, miR-۲۰۲-۳p, and miR-۴۲۹. In order to trigger the death of cells in the Hela cell line, a solitary administration of either Coumarin or Oxaliplatin was utilized. The implementation of Calcusyn software revealed a remarkable collaborative outcome between Coumarin and Oxaliplatin. Quantitative real-time PCR was used to examine the expression levels of the specific long non-coding RNAs (MALAT, PVT۱, and HOTAIR) and microRNAs (miR-۱۹۵, miR-۲۰۰b, miR-۲۰۲-۳p, and miR-۴۲۹). Results: The viability of Hela cells showed a gradual decrease in response to varying doses of Oxaliplatin, Coumarin, and the combined administration of Oxaliplatin and Coumarin. The cells that received both Coumarin and Oxaliplatin had lower levels of certain mRNAs that are normally regulated by the long non-coding RNAs HOTAIR, PVT۱, and MALAT, compared to the control group. The experiment showed that lncRNA targets such as miR-۱۹۵, miR-۲۰۰b, miR-۲۰۲-۳p, and miR-۴۲۹ had higher levels of expression in all the groups that received treatment, as compared to the control group. Conclusion: Our study revealed that the coupling of Coumarin and Oxaliplatin has an impact on the regulation of lncRNAs and their respective targets. This suggests that this combination could potentially be an effective treatment option for cervical cancer.
