Antitumor Activity of Hydatid Cyst Fluid Antigen B on Tumors Induced by MCF۷ Breast Cancer Cell in BALB/c Mice

Hydatid disease or cystic echinococcosis is a parasitic infection caused by tapeworms. It can cause cysts to grow in the liver and other organs. Studies on the experimental model of breast cancer have shown that hydatid cyst fluid (HCF) has antitumor activity. We aimed to investigate the effect of HCF and antigen B on MCF۷ breast tumor cells in BALB/c mice. Methods: The disease of sheep infected with hydatid cysts was collected from Urmia slaughterhouse and the liquid inside the cyst was concentrated by aspiration syringe and then by using a centrifuge at ۲۵۰۰ rpm. AgB was obtained using a boiling water bath for ۱۵ minutes and centrifuged at polyacrylamide gel electrophoresis for ۶۰ minutes with ۱۲.۵% sodium dodecyl sulfate (SDS PAGE). Mouse breast adenocarcinoma cell line (MCF۷) was cultured in an rpm medium and fetal bovine serum was added to it along with L-glutamine. Then ۱%Antibiotic mixture containing ۱۰۰ international units per milliliter of penicillin was added and incubation was done. These mice were divided into ۵ groups, the first PBS group and the second, third, fourth, and fifth groups intraperitoneally with concentrations of ۱۰۰, ۲۰۰, ۳۰۰, and ۵۰۰ μg/ml HCF and AgB in ۱۰۰ μl PBS. It was injected with naltrexone and naloxone. Help to determine the preventive effect of HCF and AgB antigens on the growth of MCF۷ mammary tumor in BALB/c mice, ۱۰ days after the inoculation of cancer cells, animals in each group in terms of tumor and tumor growth, blood was taken from one side of the mice. Cytokines were measured based on immunological tests. To determine the negative effect of HCF and AgB genes on MCF۷ breast tumor growth in BALB/c mice, ۱۰ days after the inoculation of cancer cells, animals in each group in terms of tumor and tumor growth, blood were taken from one side of the mice. Results: The results showed that in the mice that received the combination of HCF and AgB along with the adjuvant naltrexone and naloxone, the size of the tumor was significantly smaller in them and it was quite clear compared to the control group. And the higher the doses of our antigen combination, the smaller the tumor size. Also, the level of Th۱ cytokines such as il-۲ and il-۱۲ increased compared to the control group. On the other hand, the level of Th۲ cytokines showed a significant decrease. Histopathological studies showed reduced tumor volume and carcinogenesis. Antitumor activity of HCF and AgB may be related to immune responses against these antigens Our histopathological studies showed reduced tumor volume and carcinogenesis. Conclusion: This study evaluates the Antitumor activity of HCF and AgB on the MCF۷ breast cancer cell line in BALB/c mice. Th۱ cytokines are involved in the upregulation of an effective anticancer immune response. Increased levels of Th۱ cytokines (IL-۲, IL-۱۲, TNF-α, and IFN-γ) in HCF and AgB groups can indicate that T cells are switching to a type I immune response.
