Studying the expression of Hsa_circ_۰۰۷۵۳۴۱ and miR-۱۴۹-۵p in tumor tissue of breast cancer patients compared to normal tissue

Despite decades of laboratory, epidemiological, and clinical research, breast cancer incidence continues to rise, making it the leading cause of cancer-related diseases among women worldwide. Roughly one in ۲۰ people globally and one in eight in developed countries are affected by this malignancy. Circular RNA (circRNA) is a covalently closed loop found in eukaryotic cells without ۵'-۳' polarity or a polyadenylated tail, providing greater stability and protection compared to linear RNA. There is substantial evidence suggesting that circRNAs play a role in various pathological processes, such as the proliferation, invasion, and metastasis of numerous malignancies, including breast cancer. MicroRNAs (miRNAs) are a class of small non-coding RNAs (~۲۲nt) that contribute significantly to carcinogenesis regulation. In this study, we collected ۳۰ breast tissue samples from breast cancer patients and compared them with healthy samples. After RNA extraction and subsequent cDNA synthesis, specific primers were designed. We then employed the SYBR-Green real-time PCR method to investigate the expression of circular RNA Hsa_circ_۰۰۷۵۳۴۱ and miR-۱۴۹-۵p. Numerous recent studies have examined miR-۱۴۹-۵p and its association with various cancer types, including its relationship with other circRNAs. For instance, research on triple-negative breast cancer (cancer cells lacking estrogen and progesterone hormone receptors) and another study demonstrating its effect on specific drug resistance in breast cancer and the regulation of anaerobic glycolysis in this cancer type. A ۲۰۲۰ clinical study explored the association between miR-۱۴۹-۵p and Hsa_circ_۰۰۷۵۳۴۱, located on the MAPK۹ gene, in cervical cancer. This study found that the aforementioned circRNA can negatively regulate miR-۱۴۹-۵p, increasing tumorigenesis in cervical cancer and reducing affected patients' survival rates. Considering the similar origins of cervical and breast cancer, this research utilized real-time PCR analysis and examined the relationship between circRNA and miR expression with patients' clinical and demographic characteristics. The results were subsequently analyzed using SPSS software. Our study yielded significant findings regarding the role of these two factors in breast cancer and their potential implications for diagnosis and treatment.
