IntroductionKidney cancer is the۱۵th most common cancer worldwide, with a significantly higher incidence indeveloped countries. And the incidence is projected to rise in coming years. InEurope, kidney cancer was responsible for ۵۰ ۰۰۰ deaths in ۲۰۱۸.Early detection andscreening have been identified as priorities for kidney cancer research.Renal cell carcinoma(RCC) is the most common type of kidney cancer which could be mainly classifiedas kidney renal clear cell carcinoma (KIRC) and kidney renal papillary cellcarcinoma (KIRP). KIRP ranks second in terms of morbidity rate which comprised۱۰%-۱۵% of patients. Till now, there were few biomarkers could forecast theoutcomes of KIRP.In this article,SLC۳۴A۲ gene expression has been studied as a diagnostic biomarker in RCC patientswith metastasis.MethodsMicroarray analysis was performed on GSE۸۴۵۴۶in the field of RCC from the GEO database.SLC۳۴A۲ gene was selected as a gene withsignificantly increased expression after checking in the GEPIA۲ database, andthe survival analysis of this gene was also analyzed in this database.The signaling pathways that were effectivein RCC wereinvestigated through the KEGG database.Gene Ontology analysis was performed on SLC۳۴A۲gene through Enricher database.After analyzing the SNPs related to the SLC۳۴A۲gene through SIFT, miRNASNP and dbSNP databases, one SNP was selected in the۳'UTR region and one SNP was selected in the coding sequence, and its effectson the target gene were compared with the Hope database was analyzed.Protein-protein interaction of SLC۳۴A۲ genewas analyzed by STRING database, which interacted with ۱۰ other genes.ResultsAccording to microarray analysis and boxplot diagramming and survival analysis, SLC۳۴A۲ gene was identified as a genewith increased expression in RCCpatients.This gene was effective in Parathyroidhormone synthesis, secretion and action signaling pathway.SNP rs۱۲۱۰۳۴۳۲۹۴ was effective inincreasing the expression of SLC۳۴A۲ gene by disconnecting ۱۵ miRNAs in the۳'UTR region.The coding sequence SNP rs۱۹۹۶۷۳۵۳۹ wasfound to have a change at amino acid position ۳۷۰ from Isoleucine to Threonineafter checking with the Hope database.And the result was that the mutant residueis smaller than the wild-type residue.The wild-type residue is more hydrophobicthan the mutant residue.After gene ontologyanalysis, Cilium-Dependent Cell Motility biological process had similaritiesbetween SLC۳۴A۲ gene and RCC. ConclusionsAfter the bioinformatic analyzes performedon renal cancer patients with metastases, the SLC۳۴A۲ gene was selected as agene with significant increased expression, which can be considered as adiagnostic biomarker in renal cancer patients with metastases.SNPs rs۱۲۱۰۳۴۳۲۹۴ and rs۱۹۹۶۷۳۵۳۹ can beeffective in increasing SLC۳۴A۲ gene expression.
