Effect of Superoxide Dismutase Melon-Gliadin on VEGF and TGF β Blood Serum and Vitreous Retina in Hyperglycemia Rat Model

Hyperglycaemia can lead to the accumulation of oxidative stress and ROS, which can destruct the blood-retina barrier. VEGF and TGF β are markers of retinal disorders. SOD is an antioxidant that inhibits the process of ROS and protects the eye. the aim of this study was to determine the effect of oral administration of SOD Melon Gliadin supplements on VEGF and TGF-β blood serum and vitreous retina in a hyperglycaemia rat model. This study is true experimental laboratory carried out with Rattus Norvegicus Wistar Rat were given exposure intraperitoneal injection Streptozotocin (STZ) ۵۵ mg/kgbb to induce hyperglycaemia and treatment with SOD melon Gliadin ۵۰ mg/kgbb/day orally and divided into four groups, ۷ normal rat (negative control),۷ STZ rat (positive control),۷ SOD rat (comparison group), and ۷ STZ-SOD rat (treatment group).The Rat were terminated one month post treatment. The blood was taken and the rat's eyes were enucleated. VEGF and TGF β were evaluated in blood serum and vitreous retina of rat with Elisa and flow cytometry analysis. There was no significant difference VEGF blood serum, VEGF vitreous retina, and TGF β vitreous retina in four group but TGF β blood serum was significant in group STZ vs SOD p≤۰.۰۲۶ with Elisa. There was no significant difference TGF β vitreous retina, but VEGF vitreous retina with flow cytometry were significant differences in normal vs STZ p≤۰.۰۰۳,normal vs SOD p≤۰.۰۲۵, normal vs. STZ-SOD, p≤۰.۰۲۴. There was a significant difference VEGF vitreous retina in all control and treatment groups and a significant difference TGF β blood serum in the SOD and STZ groups. SOD Melon Gliadin supplement acts as an antioxidant can inhibit the process of ROS and can protect exposure hyperglycaemia in eye. These findings can be used in eye is highlight importance of this issue.
