Genetic Linkage Map of Oriental × Oriental Lily Population via AFLP and SSR Markers

Linkage maps are the framework for using genetic markers in biotechnological programs. In this experiment, two widely used markers were applied to find the loci of some momentous ornamental traits in lily flowers. A mapping population of ۱۰۰ F۱ progeny was used to construct the genetic linkage maps using the CP (out breeder full-sib family) model of JoinMap۴ software. A total of ۹۴۰ primers were tested, and the best ones were ۱۷۲ primer pairs, including ۹۶ AFLP markers and ۷۶ SSR markers used for map construction. A total of ۶۱۶ loci were scored, with ۴۶۵ loci for the AFLP marker and ۱۵۱ for the SSR marker. The whole mapped length was ۲۱۴۴.۲ cM. The overall number of mapped loci was ۱۸۹ loci, of which ۱۵۲ loci were assessed by AFLP markers and ۴۷ by SSR markers. The full length of linkage groups for the maternal map was ۸۶۱.۶ cM with ۱۲ linkage groups. The shortest and longest linkage group gaps in the maternal maps was ۲.۲ cM and ۵۵.۷ cM, respectively. The number of mapped loci in maternal genetic linkage maps was ۵۲ loci, consisting of ۴۲ for the AFLP markers and ۱۰ for the SSR markers. In paternal maps, the mapped loci were ۵۱, and the linkage group’s entire length was ۶۷۶ cM. The shortest linkage group gap for the paternal map was ۰.۱ cM, while the longest gap was ۴۷.۷ cM. In integrated plants, ۶ linkage groups were found with ۸۶ completely assessed loci. The total length of the linkage group for integrated maps was ۶۰۶.۶ cM. The shortest and longest gap of LGs was ۰.۲ cM and ۲۶.۸ cm, respectively.
